Not so much a great line as a great delivery, but the No Country For Old Men clip made me recall Tommy Lee Jones from the fugitive:

"I thought you didnt care."
"I dont... Don't tell anyone, okay?"

That movie comes with the greatest final line of a commentary.

At the end, Joe Wright sees the title of the movie and says "at-one-ment".

Dunno why, but that always cracked me up! :D
