Normally I just buy them in B&M's, but if I can't find it in the store, I will go over to Amazon.com.

I buy at my local Suncoast... because I have no credit card (or rather, none that still work!). Plus, I know all the employees and they usually hook me up with enough discounts to make it worth shopping there.
Best Buy (B&M) usually has the best prices on the release week of most titles.
Damn, I can remember when we used to get new release DVDs online for around $8 dollars. Most of those online stores didn't last a year. Where have all the coupons gone?!?? Those were the days!

As far as B&Ms go, Fry's Electronics out here on the left coast has a decent selection in their bargain bins ($7-10 per disc). They are also notorious for marking a newly released DVD down by as much as 50% on one day, then raising the price without warning for the following day. (Kubrick Boxed Set, anyone?)
Keep in mind that this recommendation only takes price into account, not the usually lackluster service and late shipments that plague Fry's.
www.dvdempire.com and www.dvdplanet.com for online purchases. Anywhere I happen to be for impulse buys locally. In Mobile, Alabama, we just got our first Best Buy store, so I imagine it will be getting some of my money.
Sometimes, I buy stuff on eBay, like the Alfred Hitchcock Presents and Stanley Kubrick: A Life in Pictures DVDs. These were not sold unbundled through "official" channels. I wanted these discs, but buying the boxed sets in which they were offered would have meant duplicating my efforts.
Oh... sometimes I buy previously viewed DVDs at Blockbuster or on used CD shops.

I have a Tower Video right nearby. Their prices are always comparable with Best Buy, plus there's a really cute girl there who runs the video department.

I mostly order from deepdiscountdvd.
Amazon is only good when they have a coupon AND their price is close to other sites.
Digitaleyes is good sometimes esp. if you have bonus points to get 5 off.
Always check www.dvdpricesearch.com before any ordering is done.
B&M's = brick & mortar, which means retail outlets, not online merchants.
I purchase the majority of my titles online at DVD Empire or Deep Discount DVD. Those few titles that I pick up at B&M's are usually from Best Buy.

Lately, I've been buying a bunch from a local pawn shop. They always seem to have new releases about 4-6 days after release, and they're only $10 bucks apiece.
I also buy from HW video(because I work there), and generally buy from the local guys who have the lowest price.
