Does the bracing for a subwoofer enclosure always have to be wood-based? I'm designing a 1 cubic foot enclosure using 1" thick MDF, and I was wondering if metal wall brackets or something similar might be a viable alternative(to save space inside).

No, though I'm not sure what brackets you're referring to. They must be rigid though, like the 2x2 alum angles sold at Home Depot, etc., or you could go to a metal fabricator and buy some steel angle scrap.
