Yes, you should call her back and find out "Joe's" last name. You should, however, also find out if Joe's last name is "Schmoe" (if Joe really does exist and is actually the person you should send your resume to).

Address it to whatever name you want, but if this is a larger company with an administrator/receptionist that opens and sorts the mail, it will end up at recruitment/human resources regardless of who you addressed it to.

I would save yourself the stress and simply send it general delivery to HR. That's where it will go anyway.
