Any of Wes Anderson's three flicks are great:

Bottle Rocket (hopes of a Criterion Collection seem to be fading)
Rushmore (great Criterion Collection edition)
Royal Tenenbaums (good Criterion Collection edition)

This is Spinal Tap
Waiting for Guffman
Fear of a Black Hat

Some good dark comedies by Scorsese:

After Hours
King of Comedy

For some good monologues by Spalding Gray:

Gray's Anatomy (not sure if available on DVD)
Swimming to Cambodia

Some classics:

My Man Godfrey
The Lady Eve
Sullivan's Travels
Trouble in Paradise
The Thin Man

Lots of good Marx brothers flicks but most aren't available on DVD.

Some good recommendations in this thread, as well as some real groaners in my opinion. Just goes to show how subjective movies are in general and comedy more specifically.

A few that I can never tire of and never manage to keep a straight face throughout (not that I've ever really tried); don't believe any of these have been mentioned above:

See No Evil, Hear No Evil
The Best of Abbott and Costello Live (VHS)
Gallagher: The Bookkeeper (VHS)

If those last two ever show up on DVD, I'd be a very happy camper...
