I'm not quite sure what you're asking about making reformatting two internals to share as one. But, you can move the Time Machine backup from the 250 GB drive to the new 750 GB drive, then tell Time Machine to use the 750 GB drive. It will recognize the current backups and continue as if nothing happened.

You want to use SuperDuper! to do the data move. You can't do a regular file copy with Finder.

I did essentially this to repartition my wife's Time Machine drive and preserve the existing backups.
